The Human CD200 ELISA quantitates Hu CD200 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu CD200. Principle of the method The Human CD200 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. CD200 ist ein Typ-1-Membranglykoprotein, das zwei Immunglobulin-Domänen enthält und zur Immunglobulin-Superfamilie gehört. CD200 ist ein weit verbreitetes Glykoprotein auf der Zelloberfläche, das mit einem Rezeptor auf myeloischen Zellen (CD200R) interagiert und an der lokalen Prävention der Makrophagen-Aktivierung beteiligt ist. Entfernte, aber erkennbare Homologe von CD200 wurden in vielen Herpesviren und Poxviren identifiziert. Studien der verwandten Gene in Maus und Ratte deuten darauf hin, dass CD200 die myeloische Zellaktivität reguliert und ein Hemmsignal für die Makrophagen-Linie in verschiedenen Geweben liefert. Mehrere alternativ gespleißte Transkriptvarianten wurden gefunden, die verschiedene Isoformen von CD200 codieren. Zu den Erkrankungen, die mit CD200-Dysfunktion verbunden sind, gehören das Mantellzelllymphom und die primäre hypertrophe Osteoarthropathie.
