The Rat Brain-Derived Neurotrophic Factor (Rt BDNF) ELISA quantitates Rt BDNF in rat serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Rt BDNF. Principle of the method The Rat BDNF solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Der Wachstumfaktor BDNF ist ein Mitglied der Neurotrophin-Familie. BDNF wird als pre-proBDNF synthetisiert, gefolgt von der Spaltung zu proBDNF. Obwohl die weitere Verarbeitung das reife 14 kDa-Protein erzeugt, ist pro-BDNF biologisch aktiv und wird zusammen mit der reifen Form in synaptischen Vesikeln sezerniert. BDNF wird weitgehend im Zentralnervensystem exprimiert und wirkt auf mehrere Klassen von Neuronen in autokriner und parakriner Weise. Die Signalisierung erfolgt hauptsächlich über den Tyrosinkinase-Rezeptor TrkB, obwohl auch eine Bindung an den Niederaffinitätsrezeptor p75-NTR nachgewiesen wurde. BDNF fördert das neuronale Überleben und die Differenzierung und hat sich als wichtiger Faktor bei der Gedächtnisbildung und der synaptischen Regulation erwiesen. BDNF wird von kortikalen Neuronen induziert und ist für das Überleben von Striatalneuronen im Gehirn notwendig. Die Expression von BDNF ist sowohl bei Alzheimer- als auch bei Huntington-Patienten reduziert und kann auch bei der Regulierung von Stressreaktionen und in der Biologie von Stimmungsstörungen eine Rolle spielen.
